Cofilin is an actin-binding protein that promotes the severing and depolymerization of filamentous actin (F-actin), facilitating actin filament turnover essential for processes like cell motility, cytokinesis, and endocytosis. Phosphorylation at Ser3, primarily mediated by LIM kinases (LIMK) and testicular protein kinases (TESK), inhibits cofilin's ability to bind actin, thereby reducing its actin-severing activity and stabilizing actin filaments. This regulatory mechanism allows cells to modulate actin dynamics in response to various stimuli, impacting cellular processes such as migration, morphogenesis, and response to stress. Dysregulation of cofilin phosphorylation has been implicated in pathological conditions, including cancer metastasis and neurodegenerative diseases. Detection of Phospho-Cofilin (Ser3) levels is commonly employed to assess the activation state of cofilin and the dynamics of actin remodeling in cells.
