Protein A的其他非结合域会和非目标蛋白结合，这样被洗脱下来的蛋白质纯度不够，会影响到后续的试验。 为了解决这些问题，科学家们运用基因工程技术，克隆出Protein A的基因，并对其进行改造，除去了一些不重要的非结合域。偶联这种重组Protein A的琼脂糖凝胶柱在蛋白质纯化中，的确是提高了产物的纯度。目前，市场上绝大部分重组Protein A柱都是这种产品。但是，纯化时所用的洗脱液一般为pH=2.7的甘氨酸溶液，如果洗脱效果不是很理想，还要降低pH，采用pH=1.9的甘氨酸溶液。由此可见，此法
