| In Vitro | 牛血清白蛋白(BSA)是由三个同源的全α结构域组成的583残基蛋白,以心形结构组织。牛血清白蛋白构成ca.所有血浆蛋白的60%并且结合并运输大量生理和非生理配体。牛血清白蛋白含有17个二硫键和一个未配对的半胱氨酸(Cys34),其促进二聚化并且还影响高级缔合,因为如果Cys34与另一种化合物共价结合,则聚集速率减慢。用于原纤维细胞毒性的MTT测定显示牛血清白蛋白有利于细胞生长,而不管其聚集状态。通过膜透化测定证实缺乏细胞毒性。在随后的40小时中,观察到处理细胞的存活率增加300-400%,表明与牛血清白蛋白一起孵育的细胞比未处理的细胞具有更高的存活率[1]。 |
| 细胞实验 | 在96孔微量培养板中制备样品,并且所有不含样品的孔用200μLPBS填充以使蒸发最小化。向每个孔加载100μLSH-SY5Y生长培养基,含或不含10.000 SH-SY5Y神经母细胞瘤细胞。通过添加25μL缓冲液开始测定,其中有或没有5mg/mL超声处理的牛血清白蛋白,其预先在70℃温育0,2或96小时(给出最终BSA浓度为1mg/mL)。将所有样品组合装入三联体中,并且对于六种牛血清白蛋白组合中的每一种,加载相应的三联体而不含细胞,用作空白。一个三联体也没有细胞和BSA,只有一个没有牛血清白蛋白[1]。 |

The samples were blocked with 3% bovine serum albumin (BSA) (Solarbio) and probed overnight with primary antibodies against CMTM6 (1:1000), c-MYC (1:2000) and β-actin(1:5000), and with secondary antibody horseradish peroxidase-labelled IgG (1:8000) for 2 h.

Cell(Neutrophils,5 % BSA for 1 h)
Neutrophils were fixed with 4 % parafor-maldehyde for 20 min,washed twice with PBS, and permeabilized with 0.1 % Triton X-100 for15 min. Neutrophils were then blocked with 5 % BSA for 1 h and incubated with anti-MPO antibody (1:100 dilution) before being incubated overnight with anti-CitH3 antibody (1:100 dilution) at 4℃.

Cell(ESCA cells,5 % bovine serum albumin (BSA))
Cells on the coverslips after treatments were fixed by 4 % parafor-maldehyde for 15 min, permeabilized using 0.3 % Triton X-100(Solarbio, Beijing, China) for 10 min, and blocked in 5 % bovine serum albumin (BSA) (Solarbio, Beijing, China) in PBS for 45 min. After careful washing with PBS, the cells were incubated with primary antibodies against γ-H2AX (1:200 dilutions), p53-binding protein 1 (53BP1) (1:200 dilutions), USP14(1:200 dilutions) and YAP1 (1:200 dilutions) at 4?C overnight. After rinsing with PBS, the cells were incubated with a fluorescence-conjugated secondary antibodies, including Alexa Fluor 647-Labeled Goat Anti-Mouse IgG (1:200 dilutions) and Alexa Fluor 488-Labeled Goat Anti-Rabbit IgG (1:200 dilutions). After nuclear staining with DAPI (Solarbio, Beijing, China), images were captured using a confocal laser-scanning microscope . Fluorescence density was quantified by use of the Image J 1.48 V software.
