    背景说明Diphenylterazine是一种腔肠素类似物和生物发光剂,Diphenylterazine本身仅产生很少的背景,从而形成极好的信噪比。
    生物活性Diphenylterazine (DTZ) is a bioluminescence agent. Diphenylterazine alone yielded very little background, leading to excellent signal-to-background ratios[1].
    In VitroWe use these scaffolds to design artificial luciferases that selectively catalyse the oxidative chemiluminescence of the synthetic luciferin substrates diphenylterazine3 and 2-deoxycoelenterazine. The designed active sites position an arginine guanidinium group adjacent to an anion that develops during the reaction in a binding pocket with high shape complementarity.For both luciferin substrates, we obtain designed luciferases with high selectivity; the most active of these is a small (13.9 kDa) and thermostable (with a melting temperature higher than 95 °C) Enzyme that has a catalytic efficiency on diphenylterazine (kcat/Km = 106 M-1 s-1) comparable to that of native luciferases, but a much higher substrate specificity.[2]。
    细胞实验We compared our LumiLuc - 8pyDTZ pair with several benchmark reporters in a tumor xenograft mouse model. Our new pair, which does not need organic cosolvents for in vivo administration, surpasses other reporters by detecting early tumors.LumiScarlet - 8pyDTZ was comparable to Akaluc - AkaLumine, the brightest ATP - dependent luciferase - luciferin pair, for detecting cells in deep tissues of mice.[4]。
    细胞实验BALB/c mice are used and transfected with cells expressing teLuc, Antares, Antares2 and FLuc by injecting cells into the tail vein of BALB/c mice. After the diminishing of the FLuc bioluminescence, 0.3 μmol Diphenylterazine or furimazine is intraperitoneally injected. Mice are imaged with a 1-min exposure per frame over a course of 20 min. The images are processed using the Fiji image analysis software and the frames with highest signals in individual experiments are used for comparison[1].
