The assay uses a caspase-4-specific fluorogenic substrate, Ac-LEVD-AFC. Caspase-4 cleaves this substrate generating free AFC, which can be easily quantified using a fluorescence plate reader at excitation and emission wavelengths of 400 and 505 nm, respectively.
