In Vivo
GhGLU18
Different substrates including laminarin (β-1, 3-linked polysaccharide,), laminaribiose (β-1, 3-linked disaccharide,), lichenin (β-1, 3;1, 4-linked polysaccharide, Solarbio, Beijing), pustulan (β-1, 6-linked polysaccharide, Solarbio, Beijing), and sodium cellulosate (sodium salt of β-1, 4-linked cellulose, Solarbio, Beijing) were used to test the enzyme activity of GhGLU18. GST protein and boiled GST-GhGLU18 were used as the negative controls, respectively. The standard reaction system included 10 μg GhGLU18 recombinant protein dissolved in PBS buffer and 4 % (w/v) of β-glucan substrate dissolved in 200 μL sodium acetate (100 mM, pH 5.2). The entire reaction system was first incubated at 55°C for 10 minutes, then 500 μL DNS reagent was added and incubated at 100°C for 5 minutes.
