检测原理
羊Goat growth hormone releasing hormone,GHRH ELISA KitELISA试剂盒采用双抗体夹心ELISA法检测羊样本中的Goat growth hormone releasing hormone含量。微孔板预包被抗羊Goat growth hormone releasing hormone捕获抗体，加入标准品或样本后，Goat growth hormone releasing hormone与捕获抗体结合；再加入HRP标记的检测抗体，形成抗体-Goat growth hormone releasing hormone-酶标抗体复合物；加入TMB底物显色，终止反应后测定吸光度值。吸光度值与样本中Goat growth hormone releasing hormone浓度呈正相关，通过标准曲线计算待测样本中羊Goat growth hormone releasing hormone的含量。操作步骤
1. 准备：将羊Goat growth hormone releasing hormone,GHRH ELISA KitELISA试剂盒各组分恢复至室温(18-25℃)，按需配制洗涤液工作液。2. 加样：分别在标准品孔和样本孔中加入100μL标准品或待测样本，37℃孵育90分钟。
3. 洗板：甩去孔内液体，每孔加入洗涤液，静置后甩去，重复5次。
4. 加检测抗体：每孔加入100μL HRP标记检测抗体工作液，37℃孵育60分钟。
5. 洗板：重复洗涤5次。
6. 显色：每孔加入TMB底物溶液100μL，37℃避光孵育15-20分钟。
7. 终止：每孔加入终止液50μL，轻轻混匀。
8. 测定：在450nm波长下测定各孔吸光度值(OD值)。
样本制备
血清样本：采集羊全血，室温静置30分钟后3000rpm离心10分钟，取上清液检测。血浆样本：使用EDTA抗凝采血管采集羊全血，3000rpm离心10分钟，取上清液。
组织匀浆样本：取羊组织加入PBS匀浆，4℃离心取上清液检测。
细胞上清液：收集细胞培养上清液，离心去除细胞碎片后取上清检测。
样本若不及时检测，请分装后-20℃冻存，避免反复冻融。
保存条件
