Background information
CD45 MicroBeads were developed for the direct isolation of cells expressing rat CD45 antigen, also known as leukocyte common antigen (LCA), which is a 180–220 kDa type I transmembrane protein with multiple isoforms and differential glycosylation. It appears in at least five isoforms depending on the differentiation status of the cell. CD45 is expressed at high levels on all cells of hematopoietic origin except for erythrocytes and platelets. It plays a role in signal transduction through B and T cell antigen receptors.

CD45+ cells were isolated from a rat spleen cell suspension using CD45 MicroBeads, rat a MidiMACS Separator and an LS Column. The cells are fluorescently stained with CD45-APC (# 130-107-790) and analyzed by flow cytometry using the MACSQuant Analyzer. Cell debris and dead cells were excluded from the analysis based on scatter signals and PI fluorescence.
