p21 (also known as Cyclin-dependent kinase inhibitor 1, or CDKN1A), encoded by the CDKN1A gene, is a critical regulator of the cell cycle. It acts as an inhibitor of cyclin-dependent kinases (CDKs), particularly CDK2 and CDK4, preventing the progression of the cell cycle from the G1 phase to the S phase. p21 is activated in response to various cellular stresses, including DNA damage, and is often regulated by the tumor suppressor protein p53. When p53 detects DNA damage, it induces the expression of p21, which subsequently halts cell cycle progression, allowing time for DNA repair. If the damage is irreparable, p21 can promote cellular senescence or apoptosis. Because of its role in controlling cell proliferation and maintaining genomic stability, p21 is involved in cancer prevention and is often studied in the context of tumor suppression and cellular response to stress.
