Collagen VII is a crucial structural protein in the extracellular matrix (ECM) and primarily functions as a component of the anchoring fibrils in the skin, where it plays a vital role in connecting the epidermis to the underlying dermis. It is produced by keratinocytes and fibroblasts and consists of a triple-helical structure that is key to the stability and integrity of the basement membrane. Collagen VII is essential for skin architecture, as it provides mechanical strength and stability to the dermo-epidermal junction (DEJ). This protein is heavily studied in the context of dermatology, especially in relation to diseases such as dystrophic epidermolysis bullosa (DEB), a genetic disorder characterized by skin fragility and blistering caused by mutations in the COL7A1 gene, which encodes Collagen VII. In addition to its role in skin health, Collagen VII also plays a role in tissue repair, wound healing, and other connective tissue diseases. Its function and degradation are also of interest in cancer metastasis and fibrotic conditions, where remodeling of the ECM occurs.
