In Vitro

Agrobacterium tumefaciens (GV3101)（50 mg/ ml rifampin，28℃）

Positive transformants of A. tumefaciens harboring the reporter vector (ProAcSWEET9b:Hap1::LUC or ProAcSWEET9b:Hap2::LUC, with the pGreenII-0800-LUC empty vector as a negative control) were cultured on liquid medium supplemented with 50 mg/ ml kanamycin and rifampin (Solarbio) and incubated at 28℃until they reached an OD600 of 0.6 . After centrifugation at 12 000 g for 5 min at room temperature, the positive bacterial precipitates were resuspended in infection buffer (10 mM MgCl₂,  10 mM MES,  100 mM acetosyringone) to an OD600 of 0.8, then incubated at room temperature in the dark without shaking for 2 h before infiltration.
