背景资料：BMPs (bone morphogenetic proteins) belong to the TGF beta superfamily of structurally related signaling proteins. Members of this superfamily are widely represented throughout the animal kingdom and have been implicated in a variety of developmental processes. Proteins of the TGF beta superfamily are disulfide-linked dimers composed of two 12-15 kDa polypeptide chains. As implied by their name, BMPs initiate, promote and regulate bone development, growth, remodeling and repair. Smad1 translocation to the nucleus is observed after the addition of BMP4 (also designated BMP2B), suggesting that BMP4 may play a role in activation of the Smad pathway. BMP is secreted into the extracellular matrix.

一、材料和试剂1、玻片：须用免疫组化专用玻片，或用2%多聚赖氨酸包被处理玻片。2、5-10%正常山羊血清封闭液，用PBS配。3、3%牛血清白蛋白（BSA），用PBS配。4、0.01M pH7.4 PBS5、1% BSA-PBS（含0.05% Tween20）6、AEC底物（1）底物缓冲液（20X） PH 4.6醋酸（分子量60.6） 30.6mlTriton X-100醋酸钠 3H2O（分子量136.1）66.68克加蒸馏水到960ml，调pH到4.6，最后加蒸馏水到总体积1000ml
