胶原酶Ⅳ包含至少7种蛋白酶成分，分子量从68-130KD不等。它能消化多种组织。胶原酶的使 用浓度一般为0.1- 5 mg/ml，消化过程中可以搅拌以促进酶的消化。胶原酶在消化的过程中可以引起 细胞的少量死亡，一般消化时间为15min至数小时。如果浓度发生变化那么消化时间可能更长。消 化过程中首.选的缓冲液为含钙离子和BSA的Krebs Ringer Buffer。在消化的过程中如果细胞大量死亡 则需要降低酶量，同时可以加入0.5%BSA或5-10%血清来稳定细胞的状态和消化效果。 胶原酶的灭菌首先离心或者用0.8µm的滤器去掉不溶物。然后再用0.2µm的滤器过滤除菌。盐酸 苯甲脒和TLCK联合作用可以与该酶成分中大部分的蛋白酶结合而抑制其活性，但是他们却不会抑 制胶原酶的活性。其中盐酸苯甲脒的浓度一般为1 mM，TLCK 的浓度一般为0.100-0.135 mM。值得 注意的是两者不能完全保证所有蛋白酶的活性均被抑制。
酶的溶解性：0.05 - 0.1 mg/ml 胶原酶溶于50 mM TES buffer，pH 7.4 (37 ℃) 含 0.36 mM 氯化 钙。（Cat:G0150)反应体系的终浓度为50 mM TES，0.36 mM 氯化钙，25 mg 胶原，0.005-0.01 mg 胶原酶。
酶液在含有 0.3-0.5 mM 钙离子 PH 7.5 的条件下可以在 37 ℃维持活性 5 小时。在-20℃可以维 持数月。
Collagenase IV contains at least 7 protease components with molecular weights ranging from 68-130KD. It can digest various tissues. The usage concentration of collagenase is generally 0.1-5 mg/ml, and stirring can be used during the digestion process to promote enzyme digestion. Collagenase can cause a small amount of cell death during the digestion process, usually lasting for 15 minutes to several hours. If the concentration changes, the digestion time may be longer. The first buffer selected during the digestion process is Krebs Ringer Buffer containing calcium ions and BSA. If a large number of cells die during the digestion process, it is necessary to reduce the enzyme content, and 0.5% BSA or 5-10% serum can be added to stabilize the cell state and digestion effect. The sterilization of collagenase begins with centrifugation or removal of insoluble substances using a 0.8 µ m filter. Then use a 0.2 µ m filter to filter and remove bacteria. The combination of benzamidine hydrochloride and TLCK can bind to most of the proteases in this enzyme component to inhibit its activity, but they do not inhibit the activity of collagenase. The concentration of benzamidine hydrochloride is generally 1 mM, and the concentration of TLCK is generally 0.100-0.135 mM. It is worth noting that both cannot fully guarantee that the activity of all proteases is inhibited.
Enzyme solubility: 0.05-0.1 mg/ml collagenase is soluble in 50 mM TES buffer, pH 7.4 (37 ℃), containing 0.36 mM calcium chloride. The final concentration of the (Cat: G0150) reaction system is 50 mM TES, 0.36 mM calcium chloride, 25 mg collagen, and 0.005-0.01 mg collagenase.
The enzyme solution can maintain its activity for 5 hours at 37 ℃ under conditions containing 0.3-0.5 mM calcium ion pH 7.5. It can be maintained for several months at -20 ℃
