Adsorption mechanism:
The adsorption effect of macroporous resin relies on the van der Waals attraction between it and the adsorbed molecules (adsorbates), and works through its huge specific surface area for physical adsorption. Organic compounds can be separated by certain solvents based on their adsorption capacity and molecular weight to achieve different purposes such as separation, purification, impurity removal, and concentration. Macroporous adsorption resin is a separation material that combines the principles of adsorption and screening. The adsorption of macroporous adsorption resin is essentially a surface adsorption phenomenon caused by highly dispersed objects or uneven surface molecular forces. This adsorption performance is due to van der Waals gravity or the formation of hydrogen bonds; Meanwhile, due to the porous structure of the macroporous adsorption resin, it has a screening effect on substances with different molecular sizes. Through the above adsorption and screening principles, organic compounds can be separated by eluting with certain solvents on macroporous adsorption resins based on different adsorption forces and molecular weights. 大孔吸附树脂吸附技术最早用于废水处理、医药工业、化学工业、分析化学、临床检定和治疗等领域,近年来在我国已广泛用于中草药有效成分的提取、分离、纯化工作中。与中药制剂传统工艺比较,应用大孔吸附树脂技术所得提取物体积小、不吸潮、易制成外型美观的各种剂型,特别适用于颗粒剂、胶囊剂和片剂,改变了传统中药制剂的粗、黑、大现象,有利于中药制剂剂型的升级换代,促进了中药现代化研究的发展,国家中医药管理局等单位联合发布的2002~2010《医药科学技术政策》明确提出:研制开发中药动态逆流提取、超临界萃取、中药饮片浸润、大孔树脂分离等技术。
使用方法:
在运用大孔吸附树脂进行分离精制工艺时,其大致操作步骤为:大孔吸附树脂预处理——树脂上柱——药液上柱——大孔吸附树脂的解吸——大孔吸附树脂的清洗、再生。由于每一个操作单元都会影响到大孔吸附树脂的分离效果,因此对大孔吸附树脂的精制工艺和分离技术的要求就相对较高
