描述：
T4 DNA Ligase 可催化相邻 DNA 链的 5’-P 末端和3’-OH 末端以磷酸二酯键结合的反应，需 ATP 作辅酶，不仅可以催化粘性末端之间或平滑末端之间的 DNA 的连接，也可以催化 DNA 与 RNA 之间以及少数 RNA 之间的连接。可
应用于在粘性末端或平末端连接双链 DNA 分子，也可应用于修补双链 DNA、RNA 或 DNA RNA 杂交体上的缺口。
活性定义：16°C 反应 30 分钟条件下，使 1pmol 的粘性末端
DNA 连接所需要的酶量定义为 1Unit。
失活：65°C，10 分钟。
10×T4 Ligase Buffer：500 mM Tris-HCl, pH 7.5, 50 mM
MgCl2, 25mM DTT, 5mM ATP.
操作方法
1. 按以下组分配制反应体系
10×T4 Ligase Buffer 2 μl
载体 50 ng(0.2-1pmol)
插入片段 1-50 ng(0.2-10pmol)
T4 DNA Ligase(200 U/μl) 1 μl
ddH2O Up to 20 μl
注意：两个片段的连接时，通常载体与插入片段的摩尔比为
1:3（但该比例并非绝对严谨，在 1:1-1：10 范围内均可获得理想的实验结果），提取根据片段大小与浓度，计算其摩尔浓度。
2. 如为粘末端连接反应， 22°C （16-37°C）连接 30min，
连接产物直接用于转化（或冻存于-20°C）。
