在 Western 印迹、ELISA、IHC 和其他免疫检测方案中使用 Thermo Scientific Pierce 重组蛋白 A 探测和检测兔和人源抗体（尤其是 IgG 同种型）。
重组过氧化物酶偶联蛋白 A 的特点：
•每个蛋白包含四个 Fc 结合结构域
•对于兔、猪、犬和猫血清的多克隆 IgG，优于蛋白 G
•对于小鼠 IgG1、人 IgG3 以及大鼠、山羊和牛抗体，弱于蛋白 G
蛋白 A 是由金黄色pu萄球菌的数种菌株生成的细胞壁组分，包含一个单多肽链，且仅含少量或不含碳水化合物。重组蛋白 A 在大肠杆菌中产生，其功能机制与天然蛋白 A 基本相同。蛋白 A 分子包含四个能够与多个种属（包括人和兔）IgG 中的 Fc 区相互作用的高亲和力结合位点。pH 值 8.2 时是理想结合条件，但在中性或生理条件下（pH 值 7.0 至 7.6）结合也有效。
重组蛋白 A 的属性：
• 来源：大肠杆菌
• 分子量：∼44,600（使用 SDS-PAGE 测量的表观 MW：45,000）
•形式：无盐粉末
• 0.1%溶液的 A280：∼0.165
• 等电点 (pI)：4.7-4.8
所有物种中蛋白 A 与 IgG 之间的相互作用均不等效。即使在一种物种内，蛋白 A 也与一些 IgG 亚类相互作用，而不与其他亚类相互作用。例如，与人 IgG1、IgG2 和 IgG4 结合力强，而不与 IgG3 结合。也有许多情况，单克隆抗体不与蛋白 A 结合，尤其是大多数大鼠免疫球蛋白和小鼠 IgG1。
