| 工作原理 | 本试剂盒采用的是双抗体夹心法酶联免疫吸附检测技术（ELISA）。测定样品中人α1抗胰蛋白酶水平。向预先包被了抗人α1抗胰蛋白酶抗体的酶标孔中，加入标准品和样本，温育后，加入生物素标记的抗α1抗胰蛋白酶抗体。再与HRP标记的链霉亲和素结合，形成免疫复合物，再经过温育和洗涤，去除未结合的酶，然后加入显色底物TMB，产生蓝色，并在酸的作用下转化成最终的黄色。最后，在450 nm处测定反应孔样品吸光度（OD）值，样本中的人α1抗胰蛋白酶浓度与OD值成正比，通过绘制标准曲线计算出样本中人α1抗胰蛋白酶的浓度。 |
| Purpose | This assay employs the Sandwich enzyme immunoassay technique. Determine the level of Human α1-AT(Alpha 1-Antitrypsin) in the sample. Add standard and sample to the microplate that have been pre-coated with Anti-Human α1-AT(Alpha 1-Antitrypsin) antibody. After incubation, add Biotin-conjugated anti-Human α1-AT(Alpha 1-Antitrypsin) antibody. It is then combined with HRP-conjugated streptavidin to form an immune complex, then incubated and washed to remove unbound enzyme, and then added to the chromogenic substrate TMB to produce a blue color, and converted to the final yellow under the action of acid. Finally, the absorbance (OD) value was measured at 450 nm. The concentration of Human α1-AT(Alpha 1-Antitrypsin) in the sample was proportional to the OD value. The concentration of Human α1-AT(Alpha 1-Antitrypsin) in the sample can be calculated by drawing a standard curve. |

α1-抗胰蛋白酶介绍：α1抗胰蛋白酶是一种糖蛋白。含糖10%～20%，主要由肝脏合成。它是血清中最主要的蛋白酶抑制剂，对凝血酶、尿激酶等其他酶也有抑制作用；也是一种急性时相反应蛋白，在炎症性疾患时，α1抗胰蛋白酶可透过毛细血管进入组织液，在炎症局部往往浓度很高，对急性炎性疾病有一定限制作用。 α1-抗胰蛋白酶正常值： （1）酶速率法（37℃）：0.85～2.13U/L （2）免疫比浊法：14.2～36.4μmol/L α1-抗胰蛋白酶临床意义：
α -抗胰蛋白酶（α1 -antitrypsin,α1 AT或AAT），是具有蛋白酶抑制作用的一种急性时相反应蛋白，分子量为5.5万，P1 值4.8，含有10%-12%糖。在醋酸纤维薄膜或琼脂糖电泳中泳动于α1 区带，是这一区带的主要组分。区带中的另2个主要组分；α1 -酸性糖蛋白含糖量特别高，α1 -脂蛋白含脂类特别高，因此蛋白质的染色都很浅。作为蛋白酶的抑制物，它不仅作用于胰蛋白酶，同时也作用于糜蛋白酶、尿激酶、肾素、胶原酶、弹性蛋白酶、纤溶酶和凝血酶等。AAT占血清
