520 硫醇定量检测试剂盒为硫醇浓度的测量提供了一种方便、高灵敏度的荧光检测方法。试剂盒中提供的新型试剂与硫醇反应后会产生荧光，在激发/发射=490/520 nm 时可轻松检测到该荧光。该测定的线性范围为 0.04-160 µM 硫醇。硫醇是一种含有巯基的化合物，是强大的还原剂，能够充当抗氧化剂、酶辅因子和神经调节剂。
Note: Warm all kit components until thawed to room temperature before starting the experiments.
Thiol detection reagent solution: Reconstitute one vial of Thiol detection reagent (Component A) in 100 uL of DMSO (Component D). Mix the reagents thoroughly. The stock solution is good for 1 week if stored at -20℃.
Dilute the stock solution of Thiol detection reagent 100 fold in assay buffer (Component C) according to Table 1. For each experiment, prepare fresh detection solution.
Prepare GSH standard solution: Dilute the GSH stock solution 1:50 to 200 um in assay buffer (Component C). Do 2-fold serial dilutions to get concentrations of 100, 50, 25, 12.5, 6.3, and 3.2 uM. Include a blank control.
Add 1-10 uL of test sample into microplate wells.
Note 1: Use assay buffer (Component C) to dilute test samples
Note 2: If the samples are diluted in buffers containing substances that may affect assay performance, test the same amount of that buffer with glutathione standards.
Set up the GSH standard: Add 10 uL serially diluted GSH standard solution (from Step 1.2) to the wells.
Bring the total volume of all controls and samples to 10 uL.
