胶原酶原料认证:
粗胶原酶制剂含有两种不同胶原酶的几种亚型,一种巯基蛋白酶、梭菌蛋白酶、一种胰蛋白酶样酶和一种氨基肽酶。这种胶原溶解和蛋白水解活性的组合可有效分解细胞间基质,这是组织解离的重要组成部分。该复合物的一种成分是水解酶,其优先降解天然胶原蛋白中的螺旋区域,优先在序列Pro-Y-Gly-Pro中的Y-Gly键处降解,其中Y通常是中性氨基酸。这种裂解产生易受进一步肽酶消化的产物。粗胶原酶被半胱氨酸、EDTA或邻菲咯啉等金属螯合剂抑制,但不受DFP抑制。它还被α2-巨球蛋白(一种大的血浆糖蛋白)抑制。Ca2+是酶活性所必需的。每种胶原酶的特定酶谱与获得待研究细胞的组织(或与细胞的用途)相关,并且由于相关性,Worthington已经建立了几种类型的粗胶原酶:类型1,2,3和4。
1型粗胶原酶具有胶原酶,酪蛋白酶,梭菌蛋白酶和胰蛋白酶活性的原始平衡。
2型含有较高水平的蛋白酶活性,尤其是梭菌蛋白酶。
3型含有最低水平的二级蛋白酶。
4型胰蛋白酶活性特别低,以限制对膜蛋白和受体的损伤。
5型含有较高的胶原酶和酪蛋白酶值。
6型含有高胶原酶活性,酪蛋白酶与胶原酶的比例~2:1。设计用于富集相对于 I 型胶原酶 II 型(col H)胶原酶(col G)。
7型胶原酶和酪蛋白酶活性比1型和2型胶原酶高四倍。 纯化的胶原酶,代码:CLSPA,含有最少的二级蛋白水解活性以及高胶原酶活性。
