    保存条件  Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

    产品介绍 Inositol 1,4,5-triphosphate (IP3) functions as a second messenger for a myriad of extracellular stimuli including hormones, growth factors and neurotransmitters. Receptor tyrosine kinases indirectly increase the intracellular levels of IP3 through the activation of phospholipases such as phospholipase C (PLC), which convert phosphatidylinositol-4,5 bisphosphate into IP3 and diacylglycerol (DAG). The inositol 1,4,5- triphosphate receptor, IP3R, acts as an inositol triphosphate (IP3)-gated calcium release channel in a variety of cell types. Three IP3 receptor subtypes have been described and are designated IP3R-I, IP3R-II and IP3R-III. IP3R-I is the predominant IP3R subtype expressed in neuronal tissues and the central nervous system, but is also expressed at high levels in the liver.
    Function : Intracellular channel that mediates calcium release from the endoplasmic reticulum following stimulation by inositol 1,4,5-trisphosphate.
    Subunit : Homotetramer. Interacts with TRPC4. The PPXXF motif binds HOM1, HOM2 and HOM3. Interacts with RYR1, RYR2, ITPR1, SHANK1 and SHANK3. Interacts with ERP44 in a pH-, redox state- and calcium-dependent manner which results in the inhibition the calcium channel activity. The strength of this interaction inversely correlates with calcium concentration. Part of cGMP kinase signaling complex at least composed of ACTA2/alpha-actin, CNN1/calponin H1, PLN/phospholamban, PRKG1 and ITPR1. Interacts with AHCYL1 (By similarity). Interacts with MRVI1 and CABP1 (via N-terminus).

    Subcellular Location : Endoplasmic reticulum membrane.
    Tissue Specificity : Widely expressed.
    Post-translational modifications : Phosphorylated by cAMP kinase. 
    Phosphorylation prevents the ligand-induced opening of the calcium channels.
    Phosphorylated on tyrosine residues.
    DISEASE : Defects in ITPR1 are the cause of spinocerebellar ataxia type 15 (SCA15) (SCA15) [MIM:606658]. Spinocerebellar ataxia is a clinically and genetically heterogeneous group of cerebellar disorders. Patients show progressive incoordination of gait and often poor coordination of hands, speech and eye movements, due to degeneration of the cerebellum with variable involvement of the brainstem and spinal cord. SCA15 is an autosomal dominant cerebellar ataxia (ADCA). It is very slow progressing form with a wide range of onset, ranging from childhood to adult. Most patients remain ambulatory.

      的产生及其调节(图8-16)。其中某些磷脂酰肌醇产物主要参与信号传递,如IP3为一种第二信使,引起胞浆内Ca 2+ 浓度升高,激活Ca 2+ 浓度升高,激活Ca 2+ 依赖的蛋白激酶。3,4-二磷酸磷脂酰肌醇[phosphatidylinositol3,4-bisphosphate,PtdIns(3,4)P2]及3,4,5-三磷酸磷脂酰肌醇[PtdIns(3,4,5)P3]均可以充当第二信使,在体外可激活PKCζ异型。

    • PKC系统(protein kinase C system,PKC system)

      ( diacylglycerol, DAG)和1,4,5-三磷酸肌醇(IP3)。IP3动员细胞内钙库释放Ca2 到细胞质中与钙调蛋白结合,随后参与一系列的反应;而DAG在Ca2 的协同下激活蛋白激酶C(protein kinase C,PKC),然后通过蛋白激酶C引起级联反应,进行细胞的应答, 故此将该系统称为PKC系统,或称为IP3、DAG、Ca2 信号通路。
