Protocol
The cells were fixed with 70% ice-cold methanol overnight at 4℃. Cells stained with Primary Antibody for 30 min at room temperature. The cells were then incubated in 1 X PBS/2%BSA/10% goat serum to block non-specific protein-protein interactions followed by the antibody for 15 min at room temperature. The secondary antibody used for 40 min at room temperature. Acquisition of 20,000 events was performed.

胰腺（Pancreas）是脊椎动物体内兼具消化与内分泌作用的重要器官，同时承担外分泌和内分泌两项核心功能。其主体结构为外分泌腺，主要由胰腺腺泡与导管组成，富含消化酶的胰液经由胰管被输送至十二指肠，实现对食物的分解消化。胰腺的内分泌部分则由大小不一的细胞团——胰岛构成，可分泌不同类型的激素来参与血糖水平的调控。胰岛包含多种细胞类型：分泌胰高血糖素以升高血糖的α细胞，分泌胰岛素以降低血糖的β细胞，分泌生长抑素并通过旁分泌来抑制α和β细胞分泌的δ细胞，分泌胰多肽来抑制胃肠运动、胰液分泌与胆囊
酸糖蛋白受体(asialoglycoproteins receptor，ASGPR)只存在于肝细胞表面，它能与末端为半乳糖基的蛋白质或多肽发生特异性结合；转铁蛋白受体(transferring receptor，TfR)在肿瘤细胞表面过量表达，利用转铁蛋白―聚乙烯亚胺(PEl)/DNA复合物构建成基因治疗载体，可有效转染肿瘤细胞。近年来利用抗原―抗体特异性结合也发展出了选择性基因转移方法，如利用表皮生长因子抗体可介导DNA进入表达表皮生长因子受体的细胞，由于肿瘤细胞常常过表达表皮生长因子受体，因此是一种很有希望的肿瘤靶向性基因传递方法。
