摘要

后颅窝A型室管膜瘤是一种罕见的婴儿脑肿瘤,已知体细胞突变极少,被认为由表观遗传机制驱动。PFA室管膜瘤在男性儿童中的发病率和预后均显著差于女性儿童。本研究显示,PFA室管膜瘤的细胞层级在男性个体中比女性个体分化程度更低。在正常发育的小鼠后脑中,雄性胶质祖细胞的分化程度低于匹配的雌性同胞对照。为了进一步解析染色体与性腺贡献在雄性后脑中的作用,我们使用了四核心基因型小鼠模型,结果表明是雄激素信号而非性染色体延长了雄性小鼠后脑的分化。雄激素补充促进了PFA室管膜瘤的生长,但对其他脑肿瘤无效。相反,雄激素阻断降低了PFA室管膜瘤的干细胞样潜能和增殖能力。我们得出结论,正常发育的后脑和PFA室管膜瘤中的雄激素信号足以促进生长并延迟分化。抗雄激素疗法代表了靶向这种目前无法治愈的儿童癌症的潜在临床途径。
