Rapid Coomassie Staining Procedure

Make up following solutions (maybe stored for months at room temp.) Ratios are v/v unless stated otherwise.
Stain:
40% methanol; 10% acetic acid; 50% water; 0.1 % (w/v) Coomassie Brilliant Blue R250
De-stain:
40% methanol; 10% acetic acid; 50% water; 2 Kim Wipes
- Begin staining (Procedure is for mini-gel; takes about 10 minutes to see results; use longer incubation times for thicker gels)
- Put your gel in a plastic staining container with lid
- Add enough Stain to cover gel well
- Put lid on, microwave on high for 1 minute
- Stain while rocking (up to ten minutes, strong staining is usually seen after 4 minutes; 2 minutes is good to just detect proteins)
- Pour out stain, add a large amount of de-stain, add Kim Wipes.
- De-stain on rocker at room temp.
- Should be able to see results in a few minutes after adding de-stain; leave 2 hours to overnight for complete de-staining
