Frozen Permanents
YEAST
1) grow culture to stationary in YEPD
2) Add 0.8ml of culture to 0.8ml of 30% glycerol in labelled freezer vials
3) vortex and place at -80o C
4) for strains with plasmids, pick a colony from a selective plate, grow to stationary in YEPD and freeze in glycerol. streak out on selective plates
BACTERIA
1) grow cells overnight in LB or LB plus appropriate drug
2) add 0.8ml of culture to 0.8ml of 30% glycerol in labelled freezer vial.
3) vortex and place at -80o C
