本法用于测定同一染色体上基因的排列顺序以及各个基因间的相对距离，是绘制连锁图（染色体图）的基本实验方法。例如有了对等位基因Aa，Bb，Cc作成杂合型个体，使之与三对基因均为隐性的个体进行测交，计算AB，BC以及AC间的重组值，再从这些重组值分别估计各基因间的交换值（等于图距）。设AB，BC，AC的交换值分别为x，y，z，而z=x y，则此三基因的连锁图将作如下排列：

　　当在某一连锁群中发现一个新的基因C时，可在C与同一连锁群上的2个已知基因A，B作三点试验，便能定出基因C在连锁图上的位置。
