Yeast colony hybridization is a method for determining the presence or absence of a sequence of DNA, be it of chromosomal or plasmid location, in an individual (single cell-derived) colony. It can be used, for example, to screen transformants for a nonselectable sequence or to estimate the percent of individuals in a population still containing a nonselective element like the 2-� plasmid (when estimating plasmid copy number of a nonselectable plasmid such as the 2-�, it is vital to know the percent of cells in a population without any plasmid at all; failure to take this into account can lead to misleading results).
