注意事项：
1加样：
①实验样本过多时，可分批次操作，以减少实验时间延长造成的误差；
②加酶试剂后，用吸水纸吸干孔外试剂；
③作定量试验时，使用加样器加注试剂，并经常校正其准确性，此外，要做到一份样本一个移液头，防止交叉污染。
2.温育：
①如有两种温度，尽量使用较低的温育温度、较长反应时间的条件；
②用1个小温度计放置在板孔反应液中测量观察；
③96孔板周围孔与中心孔在温育中的热力学梯度会造成“边缘效应”，因此尽量采用水浴；
3.洗板：
①手工洗板时，拍板时要垂直，避免交叉污染，用力不能过猛，防止抗原抗体复合物脱离；
②洗板机洗板时应经常检查冲洗头是否通畅，若被杂物堵塞时可用注射器针头挑出；
③为了洗涤效果好，实验室可采用机器与人工洗涤相结合的方法，在机器洗涤后再进行人工洗板1-2次，或适当增加洗板的次数；
4.显色：
ELISA试剂盒中，如以四甲基(TMB)为底物，则提供的底物为A和B两瓶应用液；如以(OPD)为底物，则试剂盒提供片剂或粉剂。显色反应条件为37℃或室温反应15-30 min。以(OPD)为底物的试剂，要临用前30 min配制且必须避光。以四甲基(TMB)为底物的试剂则不需避光，但A、B液应尽量避免接触金属器械。
5.判定：
读取结果要在15-30 min内完成，定性测定用肉眼判读试验结果时，反应板应水平距离白色背景10-15 cm；定量测定时用酶标仪读取结果，酶标仪不应置于阳光或强光照射下，需先预热15-30 min再进行测试。
操作步骤：
1）运用前，将所有试剂充沛混匀。不要使液体发生很多的泡沫，防止加样时参加很多的气泡，发生加样上的差错。
2）依据待测样品数量加上规范品的数量决议所需的板条数。每个规范品和空白孔主张做复孔。每个样品依据自个的数量来定，能运用复孔的尽量做复孔。标本用标本稀释液1：1稀释后参加50ul于反响孔内。
3）参加稀释好后的规范品50ul于反响孔、参加待测样品50ul于反响孔内。当即参加50ul的生物素标记的抗体。盖上膜板，悄悄振动混匀，37℃温育1小时。
4）甩去孔内液体，每孔加满洗刷液，振动30秒，甩去洗刷液，用吸水纸拍干。重复此操作3次。假如用洗板机洗刷，洗刷次数添加一次。
5）每孔参加80ul的亲和链酶素-HRP，悄悄振动混匀，37℃温育30分钟。
6）甩去孔内液体，每孔加满洗刷液，振动30秒，甩去洗刷液，用吸水纸拍干。重复此操作3次。假如用洗板机洗刷，洗刷次数添加一次。
7）每孔参加底物A、B各50ul，悄悄振动混匀，37℃温育10分钟。防止光照。
8）取出酶标板，敏捷参加50ul停止液，参加停止液后应当即测定成果。
9）在450nm波利益测定各孔的OD值。
