Confocal fluorescent analysis of HeLa and NIH/3T3 cells using 06-866 (Red). Actin filaments have been labeled with AlexaFluor®488-Phalloidin (Green). Nucleus is stained with DAPI (Blue). This antibody positively stains the nucleus.
Sonicated chromatin prepared from untreated HeLa cells (1 X 106 cell equivalents) was subjected to chromatin immunoprecipitation using 4 µL of either a normal rabbit antiserum or Anti-Acetyl Histone H4 serum and the Magna ChIP A (Cat. # 06-866) Kit. Successful immunoprecipitation of acetyl histone H4 associated DNA fragments was verified by qPCR using control ChIP Primers flanking the human GAPDH promoter.

Western Blot Analysis:
1:2000 dilution of this lot detected acetylated histone H4 in acid extracted proteins from HeLa cells treated with 5 mM sodium butyrate. Sodium butyrate, an inhibitor of deacetylases, was used to enhance detection of acetylated histone H4.
