To detect Human IL-17D by direct ELISA (using 100 ml/well) a concentration of approximately 1.0 μg/ml of this antibody is required. This biotinylated polyclonal antibody allows the detection of at least 0.2 – 0.4 ng/well of recombinant Human IL-17D.
To detect Human IL-17D by sandwich ELISA (using 100 ml/well) a concentration of 0.25 – 1.0 μg/ml of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech’s Polyclonal Anti- Human IL-17D (500-P88) as a capture antibody, allows the detection of at least 0.2 – 0.4 ng/well of recombinant Human IL-17D.

文献和实验上．当标记生物素的凝集素与组织细胞膜的单糖结合后，再用ABC复合物中亲和素与该生物素结合，在糖基的位置可形成一个较大的复合物，以增加检测的敏感性。其染色程序如下：1．切片脱蜡处理同前：2．小鼠肝粉(10ug/ml，PBS配制)或生物素阻断剂孵育切片?室温10分钟，抑制组织中内源性生物素；3．3％H2 O2 孵育10分钟(阻断内源性过氧化物酶，避免假阳性)，PBS漂洗2次，每次5分钟；4．用适当稀释度的生物素标记凝集素室温孵育切片45分钟：5．PBS漂洗2次，每次5分钟；6．ABC液孵育切片．
Leupeptin，终浓度为1ug/ml ⑤Pepstatin，终浓度为1uM ⑥15ul BirA酶, 终浓度为0.1uM 2. 室温下孵育过夜。 3. 将上述生物素化反应液置低温离心机中，用最大速度离心以去除沉淀。
实验试剂 硫酸葡聚糖钠盐(可不用)、聚乙烯吡赂啉酮（PVP）、聚蔗糖、牛血清白蛋白（BSA）、100mM EDTA、3MnaAC、无水乙醇、电泳缓冲液、切变性鲱鱼精DNA（100ug/ml） 20×SSC溶液 氯化钠 3M 预杂交液 6×SSC 柠檬酸三钠 0.3M （含100ug/ul变性鲱鱼精DNA） pH 7.0
