苏丹Ⅲ(Sudan Ⅲ 或 Ⅳ) 能使木栓化,角质化的细胞壁及脂肪,挥发油,树脂等染成红色或橙红色.
脂肪和苏丹染液有比较强的亲和力，苏丹3遇脂肪变橘黄色。（即萃取原理）
适合用于生物脂肪材料的鉴定，可在光学显微镜下看到被染成橘黄色的小粒。
苏丹是鉴别脂肪的染液,苏丹III(IV)可以将脂肪染成橘黄色（红色）。
