本试剂盒可测各种动物组织、培养细胞等样本中的H+K+-ATP酶活力。H＋K＋-ATPase对胃酸的分泌及胃的消化功能具有重要的生理意义，70年代Forte等人首先在牛蛙的胃粘膜上分离出一类能被K＋专一激活而不被乌本苷抑制的ATPase即H＋K＋-ATPase，随后Lee·j等人又证明了含有该酶的膜囊泡运动跨膜运输质子的作用，从而为胃酸分泌的机理奠定了分子学基础，H＋K＋-ATPase定位于胃粘膜壁细胞（Parietal lell）上，属于第二类质子泵，它通过自身的磷酸化（E1→E2）与去磷酸化（E2→E1）完成的H＋/K＋电中性（electroneutral）跨膜离子转运，不断将壁细胞内的质子运输到膜外行使泌酸的功能。
ATP酶可分解ATP生成ADP及无机磷，测定无机磷的量可判断ATP酶活力的高低。H＋K＋—ATP酶是一种能被钾专一激活而不被乌本苷抑制的ATP酶。

1、快速简便：全程约50分钟，可测100例左右样本。
2、取样量微：本法取样本量100ul。
3、稳定性好：试剂盒2～8℃存放6个月有效。
4、再现性好：变异系数CV=1.7%。
5、回收试验： X =103.3%。
6、受外界影响因素小：干扰因素少，重复性强。
7、测试面广：可测动物组织、红细胞以及各种培养细胞、各种水产等，效果均佳。
注意事项：
1. 本试剂盒的检测试剂中含有萤光素酶，反复冻融会导致其逐渐失活。尽管经测试本试剂反复冻融5次对于其检测效果无显著影响，为取得良好的使用效果，*次解冻后可适当分装保存。反复冻融过程中，可能会导致检测试剂中出现少量沉淀，此时宜平衡至室温，并尽量溶解。如仍有残留的不溶物，可以离心去除后使用，经测试不会影响后续的检测效果。
2. 检测时需使用白色或黑色的96孔板或384孔板。如果使用普通透明的96孔板或384孔板，相邻孔之间会产生相互干扰。
3. 使用说明中提供了检测ATP标准品的方法，实际检测细胞活力时通常并不需要检测ATP标准品。

本试剂盒可测各种动物组织、培养细胞等样本中的H+K+-ATP酶活力。H＋K＋-ATPase对胃酸的分泌及胃的消化功能具有重要的生理意义，70年代Forte等人首先在牛蛙的胃粘膜上分离出一类能被K＋专一激活而不被乌本苷抑制的ATPase即H＋K＋-ATPase，随后Lee·j等人又证明了含有该酶的膜囊泡运动跨膜运输质子的作用，从而为胃酸分泌的机理奠定了分子学基础，H＋K＋-ATPase定位于胃粘膜壁细胞（Parietal lell）上，属于第二类质子泵，它通过自身的磷酸化（E1→E2）与去磷酸化（E2→E1）完成的H＋/K＋电中性（electroneutral）跨膜离子转运，不断将壁细胞内的质子运输到膜外行使泌酸的功能。
ATP酶可分解ATP生成ADP及无机磷，测定无机磷的量可判断ATP酶活力的高低。H＋K＋—ATP酶是一种能被钾专一激活而不被乌本苷抑制的ATP酶。

注意事项：
1. 本试剂盒的检测试剂中含有萤光素酶，反复冻融会导致其逐渐失活。尽管经测试本试剂反复冻融5次对于其检测效果无显著影响，为取得良好的使用效果，*次解冻后可适当分装保存。反复冻融过程中，可能会导致检测试剂中出现少量沉淀，此时宜平衡至室温，并尽量溶解。如仍有残留的不溶物，可以离心去除后使用，经测试不会影响后续的检测效果。
2. 检测时需使用白色或黑色的96孔板或384孔板。如果使用普通透明的96孔板或384孔板，相邻孔之间会产生相互干扰。
