线粒体呼吸链复合物IV（细胞色素C－氧化还原酶）活性比色法定量检测试剂是一种旨在通过细胞色素C氧化酶反应系统中还原性细胞色素C 氧化后吸光峰值的降低，即采用比色法测定样品中酶活性的权威而经典的技术方法。该技术经过精心研制、成功实验证明的。其适用于各种线粒体（动物、人体、酵母等）呼吸链复合物IV 的活性检测。产品严格无菌，即到即用，活体检测，操作简捷，性能稳定。
线粒体呼吸链复合物IV （Mitochondria Complex IV），又称为细胞色素C 氧化还原酶（cytochrome coxidoreductase）或细胞色素C 氧化酶（Cytochrome c oxidase），存在于真核生物的细胞线粒体上，主要通过氧化磷酸化为细胞提供能量。呼吸链复合物IV 是由13 种不同的亚体构成的复合物，其中含有2 个血红素（heme）基团（a 和a3）和2 个铜原子，作为辅基（prosthetic groups）。呼吸链复合物IV 的遗传变异导致氧化磷酸化类疾病，包括阿茨罕默综合症和肿瘤等。基于底物还原型细胞色素C（reduced cytochrome c），受到呼吸链复合物IV 的催化，转化为氧化型细胞色素C（oxidized cytochrome c），在分光光度仪下，出现吸光值的变化（550nm 波长），由此定量测定呼吸链复合物IV 的活性。

注意事项：
1. 本试剂盒的检测试剂中含有萤光素酶，反复冻融会导致其逐渐失活。尽管经测试本试剂反复冻融5次对于其检测效果无显著影响，为取得良好的使用效果，*次解冻后可适当分装保存。反复冻融过程中，可能会导致检测试剂中出现少量沉淀，此时宜平衡至室温，并尽量溶解。如仍有残留的不溶物，可以离心去除后使用，经测试不会影响后续的检测效果。
2. 检测时需使用白色或黑色的96孔板或384孔板。如果使用普通透明的96孔板或384孔板，相邻孔之间会产生相互干扰。
3. 使用说明中提供了检测ATP标准品的方法，实际检测细胞活力时通常并不需要检测ATP标准品。

线粒体呼吸链复合物IV （Mitochondria Complex IV），又称为细胞色素C 氧化还原酶（cytochrome coxidoreductase）或细胞色素C 氧化酶（Cytochrome c oxidase），存在于真核生物的细胞线粒体上，主要通过氧化磷酸化为细胞提供能量。呼吸链复合物IV 是由13 种不同的亚体构成的复合物，其中含有2 个血红素（heme）基团（a 和a3）和2 个铜原子，作为辅基（prosthetic groups）。呼吸链复合物IV 的遗传变异导致氧化磷酸化类疾病，包括阿茨罕默综合症和肿瘤等。基于底物还原型细胞色素C（reduced cytochrome c），受到呼吸链复合物IV 的催化，转化为氧化型细胞色素C（oxidized cytochrome c），在分光光度仪下，出现吸光值的变化（550nm 波长），由此定量测定呼吸链复合物IV 的活性。

1. 本试剂盒的检测试剂中含有萤光素酶，反复冻融会导致其逐渐失活。尽管经测试本试剂反复冻融5次对于其检测效果无显著影响，为取得良好的使用效果，*次解冻后可适当分装保存。反复冻融过程中，可能会导致检测试剂中出现少量沉淀，此时宜平衡至室温，并尽量溶解。如仍有残留的不溶物，可以离心去除后使用，经测试不会影响后续的检测效果。
2. 检测时需使用白色或黑色的96孔板或384孔板。如果使用普通透明的96孔板或384孔板，相邻孔之间会产生相互干扰。
3. 使用说明中提供了检测ATP标准品的方法，实际检测细胞活力时通常并不需要检测ATP标准品。
