本试剂盒是一种利用荧光探针DCFH-DA进行活性氧检测的试剂盒。DCFH-DA本身没有荧光，可以自由穿过细胞膜，进入细胞内后，可以被细胞内的酯酶水解生成DCFH。而DCFH不能通透细胞膜，从而使探针很容易被装载到细胞内。细胞内的活性氧可以氧化无荧光的DCFH生成有荧光的DCF。检测DCF的荧光就可以知道细胞内活性氧的水平。本试剂盒本底低，灵敏度高，线性范围宽，使用方便。本试剂盒可以测定100-500个样品。
本试剂盒提供了活性氧阳性对照试剂Rosup，以便于活性氧的检测。Rosup是一种混合物(compound mixture),浓度为50mg/ml。
试剂盒组分：
DCFH-DA(10mM)——————————————0.1ml
活性氧阳性对照(Rosup, 50mg/ml) —————1ml
注意事项：
1. 探针装载后，一定要洗净残余的未进入细胞内的探针，否则会导致背景较高。
2. 探针装载完毕并洗净残余探针后，可以进行激发波长的扫描和发射波长的扫描，以确认探针的装载情况是否良好。DCF的激发光谱和发射光谱请参考下页图谱。
3. 尽量缩短探针装载后到测定所用的时间(刺激时间除外)，以减少各种可能的误差。
储存条件：-20℃，有效期一年。

活性氧检测试剂盒(荧光探针法) 使用注意事项
1． 微量试剂取用前请离心集液。
2． Annexin V-EGFP，Propidium Iodide（PI）避光保存及使用。
3． Propidium Iodide （PI）有毒，操作时请戴手套。
4． 本试剂盒适用于检测活细胞，流式细胞仪检测时，细胞数量不应低于1×105，，不推荐用于检测组织样本。
5． 推荐使用悬浮培养细胞。如果是贴壁细胞，需用不含EDTA的胰酶消化，如消化不当，可能引起假阳性，而用细胞刮子会造成细胞粘连成团，而影响检测。可将胰酶消化后细胞的保存在含2%BSA的PBS中，防止进一步的损伤。
6． 细胞固定后可能导致荧光的淬灭，请不要固定样品。
7． 因检测细胞的类型、凋亡诱导剂种类、使用的检测仪器不同，因而流式检测的荧光补偿也不同，因此建议每次检测均需使用未经凋亡诱导处理的细胞作为对照，进行荧光补偿的调节。

Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.
