| 背景介绍 | 白细胞介素17A(IL-17或者IL-17A),是IL-17家族细胞因子类群中的代表成员。IL-17细胞因子可通过增加组织中趋化因子的产量来有效召集单核细胞和中性粒细胞到炎症部位,从而延缓炎症的发生。IL-17是由辅助型T淋巴细胞产生,可被IL-23诱导。当细胞外的病原体入侵到免疫系统时,作为促炎性细胞因子的IL-17就会发生应答反应,破坏病原体细胞。IL-17可与肿瘤坏死因子和IL-1协调作用,在许多免疫相关疾病,如类风湿性关节炎、哮喘、狼疮、同种异体移植排斥、抗肿瘤免疫力、牛皮癣、多发性硬化症中发挥着重要作用。 |
| 检测原理 | 本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗小鼠IL-17A抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品和待测样本,经过孵育,样本中存在的IL-17A与固相抗体结合。洗涤去除未结合的物质后,加入生物素化的检测抗体孵育。洗涤去除未结合的生物素化的抗体,加入辣根过氧化物酶标记的链霉亲和素 (Streptavidin-HRP)。洗涤后,加入信号增强剂孵育,洗涤去除未结合的物质后,再次加入Streptavidin-HRP。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中IL-17A的浓度成正比。加入终止液终止反应,在450 nm波长(参考波长570-630 nm)测定吸光度值。 |

IL-17主要由Th17型细胞产生,是目前已发现的30余种白细胞介素中的一员,按序号排在第17位。该家族包括6个成员配体(IL-17A ~ IL-17F)和5个受体(IL-17RA ~ IL-17RD和SEF)。IL-17是一种主要由活化的T细胞产生的致炎细胞因子,可以促进T细胞的激活和刺激上皮细胞、内皮细胞、成纤维细胞产生多种细胞因子,从而导致炎症的产生。

Enables protein heterodimerization activity and protein homodimerization activity. Involved in several processes, including defense response to other organism; intestinal epithelial structure maintenance; and positive regulation of gene expression. Acts upstream of or within cellular response to interleukin-1; positive regulation of cytokine production involved in inflammatory response; and positive regulation of transcription by RNA polymerase II. Located in external side of plasma membrane and extracellular space. Human ortholog(s) of this gene implicated in Mycobacterium avium complex disease; autoimmune disease (multiple); bronchial disease (multiple); eye disease (multiple); and rhinitis (multiple). Orthologous to human IL17A (interleukin 17A).
