Product Description:Human glycogen synthase kinase ELISA Kit allows for the in vitro quantitative determination of glycogen synthase kinase concentrations in serum, plasma, tissue homogenates, cell culture supernates or other biological fluids.
ELISA Test Principle:
The microtiter plate provided in Human glycogen synthase kinase ELISA Kit has been pre-coated with an glycogen synthase kinase antibody specific to glycogen synthase kinase .Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for glycogen synthase kinase and then avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain glycogen synthase kinase, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm.The concentration of glycogen synthase kinase in the samples is then determined by comparing the O.D. of the samples to the standard curve.

使糖原合威酶（Ⅰ型）由 ATP而磷酸化，转化为依赖葡萄糖 -6-磷酸的非活性型（ D型）。此磷酸化反应，虽可在依赖环状 AMP的蛋白激酶或由 Ca2 而活化的磷酸化酶 b激酶催化下完成，但本酶的特异性强，只用 ATP仅对糖原合成酶磷酸化。存在于肌肉及肝中，与其它酶一起进行作用，但对糖原合成酶的不同的丝氨酸残基磷酸化，结果生成作用稍有不同的一些非活性型。磷酸化的非活性型，可由糖原合成酶 D磷酸酯酶的作用水解成活性型。
