T4 DNA Polymerase(T4 DNA 聚合酶) 催化以引物化的单链DNA 为模板，按5` → 3` 方向合成DNA。尽管具有很强的3` → 5`纠错外切酶活性，T4 DNA Polymerase 不具有5` → 3` 外切酶活性。T4 DNA Polymerase 可以利用标记的或未标记的dNTPs 补平5` 突出末端，或从具有3` 突出末端的DNA 分子得到平末端DNA。
