描述:线粒体/胞浆制备试剂盒 (Mitochondria Isolation Kit)用于从组织或培养细胞中分离线粒体和细胞胞浆成分。加入分离溶液,匀浆破碎组织细胞,经过数次800g和12000g离心,在60分钟内即可分离出完整的线粒体和胞浆成分。制备的线粒体具有很高的生物学活性,可进行各种功能研究如酶学测定,更可用于Western Blot、2D-胶、线粒体蛋白或DNA提取、蛋白质组学等研究。
严格按照说明操作,总是能制备获得高纯度线粒体。一篇方法学研究论文发现,用普利莱试剂盒制备线粒体的得率、活性、纯度优于蔗糖密度梯度离心法和Invotrogen/Pierce线粒体提取试剂盒方法。
适用:从组织、培养细胞制备高纯度线粒体,同时分离细胞胞浆成分。
组成:Mito Solution 100 ml for 50 次制备
200 ml for 100 次制备
储存: −20 ºC 12个月有效
操作步骤:
以下所有操作均在4 ºC进行
- 组织匀浆:100~200 mg新鲜组织如肝、脑、肾、心肌等,剪为0.5cm2碎块放入小容量玻璃匀浆器内。估计组织块总体积。加入1.5 ml冰预冷的Mito Solution。用间隙严紧的研杵上下研磨组织20次。
- 将匀浆液转移到离心管中,800 × g ,4 ºC离心5 min。(胞核、膜碎片、未裂解细胞等在管底,弃去)
- 收集上清液并转移到新的离心管。再次800 × g 离心5 min at 4 ºC,弃沉淀。
- 将上清液转移到新的离心管。10,000 × g 离心10 min 4 ºC。线粒体沉淀在管底。离心后的上清含胞浆成分,可收集用于对照实验。
- 洗涤线粒体:加入0.2 ml Mito Solution,轻弹管底重悬线粒体沉淀;12,000 × g 离心10 min 4 ºC。弃上清,线粒体沉淀在管底。
- 重悬线粒体:可以用Mito Solution重悬线粒体沉淀,也可以用合适后续实验的自备缓冲液来裂解线粒体沉淀,具体用量是:约每100 mg组织提取的线粒体用50 µl,约每5 × 107个细胞提取的线粒体用100 µl。用量请根据组织或细胞类型进行微调。
- 裂解线粒体后,进行蛋白浓度测定。立即使用或−70 ºC保存。
说明:
- 制备高质量线粒体的关键:第一,全程低温操作,将样品管放在冰水浴而不是碎冰块中;第二,快速,微量制备比大规模制备操作更快速,更容易获得完整的线粒体,且得率高;第三,在不破坏亚细胞器的情况下破碎细胞是制备线粒体的最关键环节。破碎效果与组织细胞类型有关;与组织块相比,贴壁培养细胞在用玻璃匀浆器匀浆时较难破壁。用1-3 ml小容量玻璃匀浆器(而不是其它破碎装置)上下研磨培养细胞20-30次。玻璃匀浆器须配套选用间隙严密的研杵,其特征是将研杵插入匀浆器套管后,可提起研杵而套管不会脱落。正确的匀浆不是旋转研杵,而是上下缓慢推拉研杵,研杵推进和提升过程中细胞遭受压力的剧变而破碎。研磨程度可用相差显微镜进行检查当未裂解细胞在~50%即可。过度研磨会破坏线粒体,而研磨不足将降低线粒体得率。
- 如果不得不用电动匀浆器(polytron),可选用6500转,刀头上下进出4次共计10秒。不同的电动匀浆器性能差别甚大,除非步步监测,否则分离结果难以预料。
- 不同的离心机必须跟据离心力g计算正确的离心转速(允许10%波动),否则将导致不纯。
- 进行Western Blot可直接用RIPA裂解液或SDS-PAGE样品缓冲液裂解线粒体;也可先用Mito Solution重悬线粒体,再加入2x SDS-PAGE样品缓冲液。
- Cytochrome oxidase, Monoamine oxidase, Succinate dehydrogenas, Glutamate dehydrogenase可用作线粒体的标志酶。
- 重悬线粒体:可以用Mito Solution重悬线粒体沉淀,也可以用合适后续实验的自备缓冲液来裂解线粒体沉淀具体用量是:约每100 mg组织提取的线粒体用50 µl,约每5 × 107个细胞提取的线粒体用100 µl。用量请根据组织或细胞类型进行微调。
- 裂解线粒体后,进行蛋白浓度测定。立即使用或−70 ºC保存。
