应用
胰岛素-转铁蛋白-硒添加剂(100╳ITS液体培养基添加剂)作为培养基补充剂适用于:
- α小鼠肝12细胞(AML-12)
- 大鼠胎儿睾丸外植体
- 人牙髓干细胞(DPSC)
生化/生理作用
- 胰岛素:一种多肽激素，可促进葡萄糖和氨基酸的摄取，这种特性可能与观察到的有丝分裂作用有关。
- 转铁蛋白:铁运输蛋白。
- 硒:通常由血清提供的必需微量元素。
组分:含有1.0mg/ml人重组胰岛素，0.55mg/ml人转铁蛋白(基本上不含铁)以及0.5μg/ml 100x浓度的亚(xī)酸✕(suān)钠。
其他说明:ITS是人重组胰岛素、人转铁蛋白和亚(xī)酸(suān)钠的一种混合物。它是一种通用的细胞补充剂设计用于含有丙✕酮酸钠的非复合型培养基(如MEM、RPMI-1640)及复合型培养基(如Ham?s F-12、DME/F-12、MEM)。
制备说明:使用不含酚红的Earle′平衡盐溶液(EBSS)制备。

！我们先在培养基中直接加入 37% 甲醛溶液 0.27 ml，使终浓度达到 1%（因为当时我们每盘细胞的培养基都是 10 ml，所以加入的甲醛溶液体积为 10 ml/37=~0.27 ml）。加入甲醛后，立刻将培养皿在摇床上室温缓慢摇动 1 分钟（确保彻底摇匀甲醛），然后静置 9 分钟。接着，迅速在每个盘子里加入 1 ml 10X 甘氨酸溶液，在摇床上室温缓慢摇动孵育 5 分钟，以终止上述固定反应，这个时候你会看到培养基变色了。由于我们使用的是贴壁细胞，所以接下来直接弃去每个盘子里的上清，然后迅速
