    Propagation: ATCC complete growth medium: The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
    Atmosphere: air, 95%; carbon dioxide (CO2), 5%
    Temperature: 37.0°C
    Subculturing: Protocol:
    1. Remove and discard culture medium.
    2. Briefly rinse the cell layer with 0.25% Trypsin - 0.53 mM EDTA solution to remove all traces of serum which contains trypsin inhibitor.
    3. Add 2.0 to 3.0 ml of Trypsin-EDTA solution to flask and observe cells under an inverted microscope until cell layer is dispersed (usually within 5 to 15 minutes).
      Note: To avoid clumping do not agitate the cells by hitting or shaking the flask while waiting for the cells to detach. Cells that are difficult to detach may be placed at 37C to facilitate dispersal.
    4. Add 6.0 to 8.0 ml of complete growth medium and aspirate cells by gently pipetting.
    5. Add appropriate aliquots of the cell suspension to new culture vessels.
    6. Incubate cultures at 37C.

    Subcultivation Ratio: 1:4 to 1:8 is recommended
    Medium Renewal: Every 2 to3 days
    Preservation: Freeze medium: Culture medium, 95%; DMSO, 5%

      都是贴壁细胞,在消化传代过程中,步骤如下:倒尽旧的培养液->用无血清的培养基清洗一两次->加入一定量的胰酶,置于37度培养箱中5--10分钟,使细胞悬浮->显微镜下观察,待细胞大部分变圆时,回到超静台->加入一定量的含血清的新培养液,以终止胰酶作用->反复吹打细胞->再置显微镜下观察,直到细胞全部悬浮起来->吸出一部分加入新的培养瓶中->最后再补充加入一定量新的培养液。注意: 1、吹细胞时尽量多吹边角儿,此处细胞生长的多。2、吸出细胞前要混匀,可以剧烈震荡培养瓶。3、我们用的是DMEM
