概述:
Golden Gate 可高效、无痕的克隆组装 DNA 片段，该方法起始于 1996 年，这是第一次使用 IIS 型限制性内切酶的和 T4 连接酶顺序或同时的活性将多个目的基因插入到同一个载体骨架上。这类核酸内切酶产生用户确定唯一的末端序列，该内切酶在识别位点之外进行切割。产生可连接的 DNA 突出末端，临近的互补 DNA 末端退火。一旦组装上，结构中内切酶的识别位点已经不存在。因为每一个互补可连接的 4 碱基突出端都是钝端（内切酶识别位点已经不存在），目的组装产物随时间逐渐积累。特别适用与多重组装实验，例如 TALEs （Transcription Activator Like Effectors）（5）和 TALENs（TALEs fused to a FokI nuclease catalytic domain）（6）。Golden Gate 方法也适用于单一目的基因的克隆实验。
