溶液的配制:
标准品:10mg无水葡萄糖(干燥失重<0.2%)。临用前加入1mL蒸馏水溶解,配制成10mg/mL葡萄糖溶液备用,2-8℃保存两周,或者用饱和苯甲酸溶液溶解,可保存更长时间。
产品说明:
CL(EC 3.2.1.4)存在于细菌、真菌和动物体内,能够催化纤维素降解,是一类可广泛应用于医药、食品、棉纺、环保及可再生资源利用等领域的酶制剂。
采用3,5-二硝基水杨酸法测定CL催化纤维素降解产生的还原糖的含量。
注意:实验之前建议选择2-3个预期差异大的样本做预实验。如果样本吸光值不在测量范围内建议稀释或者增加样本量进行检测。
需自备的仪器和用品:
可见分光光度计/酶标仪、水浴锅、低温离心机、可调式移液器、微量玻璃比色皿/96孔板、研钵/匀浆器、冰和蒸馏水。
操作步骤:具体操作步骤详见“产品资料”附件
一、样本处理(可适当调整待测样本量,具体比例可以参考文献)
- 细菌或细胞:收集细菌或细胞到离心管内,离心后弃上清;按照每500万细菌或细胞加入1mL提取液,超声冰浴破碎细菌或细胞(功率200w,超声3秒,间隔10秒,重复30次);8000g 4℃离心10min,取上清,置冰上待测。
- 组织:称取约0.1g组织加入1mL提取液,冰浴中匀浆。8000g ,4℃离心10min,取上清,置冰上待测。

(一)检测原理 血细胞比容(Hct、Ht、HCT或PCV)是指在一定条件下,经离心沉淀压紧的红细胞在全血样本中所占比值。 1、离心法:包括温氏法(Wintrobe法)、微量法:是将抗凝血置于孔径统一的温氏管或毛细玻管中,以一定转速离心一定时间后,计算红细胞层占全血的体积比。 2、血液分析仪法:原理是当细胞通过计数小孔时,形成相应大小的脉冲,脉冲的多少即为细胞数量,脉冲高度为细胞体积,通过红细胞平均体积(MCV)和红细胞计数(RBC)即求得血细胞比容,Hct=MCV×RBC
一致。三、间接凝集的分类 1、根据载体的不同,可分为间接炭凝、间接乳胶凝集和间接血凝等。2、根据吸附物不同可分为间接凝集反应(吸附抗原)和反向间接凝集反应(吸附抗体)。3、根据反应目的不同,又可分为间接凝集抑制反应和反向间接凝集抑制反应。4、根据用量和器材的不同又可分为试管法(全量法)、凹窝板法(半微量法)和反应板法(微量法)。
1、手工法:有折射计法、粘度法、比密测定法、离心法和放射性核素法。温氏法:采用中速离心,不能完全排除红细胞间残留血浆,测定结果偏高,已属淘汰。微量法:采用高速离心,细胞间残留血浆比温氏法少(约2%),且样本用量小、操作简便、残留血浆1%~3%。 2、血液分析仪法:仪器法CV为1%,手工法CV为2%,仪器法应注意红细胞增多症或血浆渗透压异常时会出现误差。
