    背景介绍
    人血管细胞粘附分子-1(VCAM-1)是一种由 715 个氨基酸残基组成、
    大小约为 100~110 kDa 的Ⅰ型跨膜糖蛋白，主要特征是存在 7 个 C2 型免
    疫球蛋白结构域，胞外区长度为 674 个氨基酸残基，跨膜区为 22 个氨
    基酸残基，胞内区为 19 个氨基酸残基。在胞外区，有多个 N-连接的糖
    基化位点(预测分子量为 80 kDa)，每个 C2 结构域上都有一个二硫键。
    VCAM-1 的种间同源性很高，小鼠/大鼠 VCAM-1 的氨基酸序列与人类
    的同源性高达 75%，而位于胞内区的 19 个氨基酸残基序列在小鼠、大
    鼠和人类中绝对保守，表达小鼠 VCAM-1 的细胞能够同时与小鼠和人
    类的白细胞结合，这也反映了它们氨基酸序列的高度一致性。VCAM-1
    中还存在一种由 3 个 C2 型结构域组成的 43 kDa 的蛋白通过糖基磷脂酰
    肌醇（GPI）连接的形式，虽然它能够与 VCAM-1 的配体（或共受体）
    相结合，但其功能尚不清楚。
