细胞增殖及细胞毒性检测试剂盒 Cell Counting Kit-8 (CCK-8) 是应用新型的水溶性四唑盐 2-(2-甲氧基-4-硝苯基)-3-(4-硝苯基)-5-(2,4-二磺基苯)-2H-四唑单钠盐快速高灵敏度检测细胞增殖和细胞毒性的比色检测产品。
本四唑盐在电子载体存在的情况下能够被线粒体内的一些脱氢酶还原成橙黄色的水溶性的 Formazan，生成的Formazan 量与活细胞数量成正比。细胞增殖越多越快，则颜色越深；细胞毒性越大，则颜色越浅。对于同样的细胞，
颜色的深浅和细胞数量呈线性关系。
CCK-8 溶液可以直接加入到细胞样品中，不需要预配各种成分，CCK-8 溶液很稳定，对细胞没有毒性，可以长时间孵育细胞。CCK-8 法测定细胞增殖或毒性试验中活细胞数目的灵敏度高，无放射性，检测细胞增殖实验的灵敏
度比其它方法如 MTT, XTT, MTS 和 WST-1 高。
使用方法：
细胞活性、细胞增殖- 毒性检测
1、收集细胞，加细胞悬液 100ul（约 5000-10000 个细胞）到 96 孔板（边缘孔用无菌水或 PBS 填充）。每板设
对照（加 100ul 培养基）。
2、置 37℃，5% CO 2 孵育过夜，倒置显微镜下观察。
3、每孔加入 10 ul 待检测药物溶液， 37℃孵育。
4、每孔加入 10 ul CCK-8 溶液，37℃孵育 1-4 小时。
5、测定 450 nm 各孔的吸光值，如无 450nm 滤光片，可以使用 450-490nm 的滤光片。
6、同时设置空白孔（培养基和 CCK-8 溶液，无细胞），对照孔（不加药培养基和 CCK-8 溶液，有细胞），每组
设定 3-5 复孔。
细胞活力计算：
将各测试孔的 OD 值减去调零孔 OD 值或对照孔 OD 值。各重复孔的 OD 值取平均数。
细胞活力% =（（加药细胞 OD - 空白 OD）/（对照细胞 OD - 空白 OD））×100
抑制率% = （（对照细胞 OD - 加药细胞 OD）/（对照细胞 OD - 空白 OD））×100
其中： 加药细胞孔：加了待测药物的实验孔(或其他方法处理的待测细胞)
对照细胞孔：不加待测药物的细胞孔
空白孔：不含细胞和药物，只加培养基和 CCK-8 试剂
细胞数量测定
1、先用细胞计数板计数所制备的细胞悬液中的细胞数量，然后接种细胞。
2、按比例 (例如：1/2 比例) 依次用培养基等比稀释成一个细胞浓度梯度，一般要做 3-5 个细胞浓度梯度，每
组 3-6 个复孔。
3、接种后培养 2-4 小时使细胞贴壁，然后加 CCK-8 试剂培养一定时间后测定 O.D 值，制作出一条以细胞数量
为横坐标 (X轴)，O.D值为纵坐标 (Y轴) 的标准曲线。根据此标准曲线可以测定出未知样品的细胞数量 (使
用此标准曲线的前提条件是实验的条件要一致，便于确定细胞的接种数量以及加入 CCK-8 后的培养时间)。
