General description
可作用于单链RNA的嘧啶特异性核糖核酸内切酶。不含DNase的RNase是一种核糖核酸酶的异质性混合物，每并按照现有质量控制的过程进行制备以确保无脱氧核糖核酸酶的存在。不含DNase的RNase特别适合用于DNA分离过程。在使用之前，绝大多数的RNase制备必须经煮沸以去除DNase活性。RNase的制备无需煮沸；它可直接从管中进行使用。
Application
不含DNase的RNase可将来自质粒或基因组DNA制品中的RNA污染有效去除。
Unit Definition
一个Kunitz单位表示在测定条件下导致A0 to A1吸光度在1分钟内降低的酶量。A0至A1对应于总的转化，A1作为最终的吸光值。
一个单位可在260 nm吸光值上造成下降，而这相当于在+25 °C一分钟内RNA至寡核苷酸的总转化。
Physical form
溶液, 500 μg/ml, 于10 mM Tris-HCl, 5 mM CaCl2, 50% 甘油(pH 7.0)。
Preparation Note
工作浓度：不含DNase的RNase的最佳工作浓度是2至5 μg/ml。该反应体积不同于其他应用。可参考以下指导建议：
- 对于小规模的质粒DNA分离（从1.5 ml细菌培养物中的“小抽”），在50 μl.的反应体积中使用 0.5 μl的不含DNase的RNase。
• 如需从100 ml细菌培养物中进行质粒DNA的分离，可在2 ml的反应体积中使用8 μl 的不含DNase的RNase。
• 如需从培养的哺乳动物细胞中进行基因组DNA的分离（5 x107细胞），使在2 ml的反应体积中使用8 μl 的不含DNase的RNase。
