关于Nectin-1/PVRL1：
Nectin-1 is a type I transmembrane glycoprotein belonging to the Ig superfamily. Nectin-1 promotes cell-cell contacts by forming homophilic or heterophilic trans-dimers. Heterophilic interactions have been detected between Nectin-1 and Nectin-3 and between Nectin-1 and Nectin-4. Nectin ECDs contain three Ig like domains: an N terminal V type that mediates ligand binding, and two C2 type. Nectin-1 binds viral Glycoprotein D to mediate Herpesvirus (but not Poxvirus) entry into vaginal mucosa, sensory neurons and fibroblasts. In forming adherens junctions and synapses, Nectin-1 and Nectin-3 initiate cell-cell interactions, recruiting αvβ3 integrin extracellularly and cadherins intracellularly through afadin and other junctional proteins. These interactions organize the cytoskeleton, strengthen attachment to basement membrane and promote further cell-cell connections. Nectin-1 and Nectin-3 have been found to localize assymetrically along the chemical synapse, with Nectin-1 primarily on the axonal side and Nectin-3 on the dendritic side. Deficiency of Nectin-1 can result in cleft lip/palate ectodermal dysplasia. Nectin-1 downregulation in epithelial cancers is mediated in part by ectodomain shedding, but it may contribute to invasiveness.

Pentraxin 2 (PTX2), also known as Serum amyloid P (SAP), is a highly conserved, naturally circulating plasma protein and a soluble pattern recognition receptor of the innate immune system. The unique binding activities indicated that it may play an important role in the removal of damaged tissue. PTX2 belongs to the pentraxin family, is universally present in amyloid deposits. Mouse with targeted deletion of the PTX2 gene shows retarded and reduced induction of experimental reactive systemic (AA type) amyloidosis confirmed that it does indeed contribute to pathogenesis of amyloidosis and is a valid therapeutic target. In recent discovery, PTX2 can be used as a powerful antifibrotic agent to regulate certain monocyte differentiation states.

关于ST2/IL-1 R4 ：
ST2, also called IL-1 R4, is an Interleukin-1 receptor family glycoprotein that plays a role in Th2 immune responses. ST2 is expressed on the surface of mast cells, activated Th2 cells, macrophages, and cardiac myocytes. This receptor is very similar to the IL-1 receptor type I and the IL-18 receptor α chain in that ST2 also has three extracellular Ig domains and an intracellular Toll domain. ST2 binds IL-33, enhances inflammatory cytokines by activating nuclear factor-κB (NF-κB) and mitogen activated protein (MAP) kinases. ST2 exists as either a membrane bound form (ST2L) or as a soluble form (sST2). ST2L acts as a transmembrane signalling receptor for IL-33 by mediating the effect of IL-33 on the inflammatory process, while sST2 can suppress IL-33 activity.
