The protein is the receptor that involved in the costimulatory signal essential for T-lymphocyte proliferation and interleukin-2 production, by binding CD28 or CTLA-4. It may play a critical role in the early events of T-cell activation and costimulation of naive T-cells, such as deciding between immunity and anergy that is made by T-cells within 24 hours after activation. Isoform 2 interferes with the formation of CD86 clusters, and thus acts as a negative regulator of T-cell activation. The protein interacts with MARCH8， human herpesvirus 8 MIR2 protein, adenovirus subgroup B fiber proteins and acts as a receptor for these viruses.It is expressed by activated B-lymphocytes and monocytes and promoted by MARCH8 and results in endo*(代"c")ytosis and lysosomal degradation.It contains 1 Ig-like C2-type(immunoglobulin-like) domainand 1 Ig-like V-type (immunoglobulin-like) domain.

Fibronectin type III domain-containing protein 5, the precursor of irisin, is a protein that is encoded by the FNDC5 gene.Human Irisin is synthesized as a 212 amino acid (aa) precursor encoding a type 1 transmembrane protein with a 121 aa extracellular domain (ECD), a 21 aa transmembrane domain, and a 39 aa cytoplasmic domain. The ECD of Irisin contains a fibronectin type III domain and multiple glycosylation sites. The ECD is proteolytically cleaved to release the 112 aa soluble Irisin hormone into circulation.Mature human, mouse share 100% sequence identity.Irisin induces expression of peroxisome proliferatoractivated receptor γ coactivator 1α (PGC1α) and uncoupling protein1(UCP1), mitochondrialassociated metabolic proteins. Irisin induces the transition of white adipose tissue into more metabolically active beige adipose tissue.Irisin also regulates neuronal cell differentiation and neurite outgrowth in the brain and is involved in the differentiation of osteoblasts.

  本酶能作用于双链DNA，沿3"→5"方向逐步切去单核苷酸。酶与底物的每次结合，只切除最末的几个核苷酸，从而对双链DNA产生渐进缺失。最适底物是平末端或3"凹陷末端DNA，亦可作用于双链DNA的切刻产生单链gap，对于对单链DNA无活性。3"突出末端抗该酶的切割，拮抗程度随3"突出末端的长度而改变，四碱基或更长的突出完全不能被切割。这种特性对于一端是抗性位点（3"突出端）一端是敏感位点（平端或5"突出端）的线性DNA分子，可以产生单向缺失。例如，可将双链DNA用产生不同末端的限制性内切酶双酶切后，利用Exonuclease III的3"→5"的外切酶活性，从一端降解，得到互补的单链 DNA和5"-P单核苷酸。与BAL 31 Nuclease 相比，本酶的碱基特异性较小，如在富含GC的位置上，由于反应停止的机率较低，可用于DNA的Deletion制作。
  本制品是把 Exonuclease III基因 (E.coli) 在大肠杆菌中进行表达后，经多次纯化分离而得到的。

使用建议：核酸外切酶 III不能切割硫代磷酸的桥连。因此，也可以通过引入一个 硫代磷酸核苷酸将DNA分子的一端位点保护起来，再创建单向缺失。

活性定义：在37℃，30分钟条件下，催化产生1 nmol酸可溶性总脱氧核糖核苷酸所要求的酶量定义为一个活性单位。

质量保证：经多次柱纯化，SDS-PAGE胶检测仅可见清晰单一的目的条带；PCR方法检测无大肠杆菌DNA残 留，无核酸内、外切酶污染。

热失活：70℃，20分钟。
