关于Testin：
Testin belongs to the prickle/espinas/testin family. Testin contains three LIM zinc-binding domains and one PET domain, TES as a scaffold protein that may have a role in cell adhesion, cell spreading and in the reorganization of the actin cytoskeleton. In addition, TES can also as a tumor suppressor, inhibits tumor cell growth, regulate the cell proliferation.TES interacts with many cytoskeletal proteins, such as Zyxin, Mena, Actin, Talin and VASP. The ability of TES to associate with alpha-actin, Zyxin and paxillin is dependent on the conformational state of the molecule.

关于OX40/TNFRSF4：
OX40, also termed CD134 and TNFRSF4, is a T cell co-stimulatory molecule of the TNF receptor superfamily which plays a key role in the survival and homeostasis of effector and memory T cells. OX40 is expressed on CD4+ and CD8+ T cells upon engagement of the TCR by antigen presenting cells along with co-stimulation by CD40-CD40 Ligand and CD28-B7. The interaction between OX40 and OX40 ligand (OX40L) will occur when activated T cells bind to professional antigen-presenting cells (APCs). The T-cell functions, including cytokine production, expansion, and survival, are then enhanced by the OX40 costimulatory signals. OX40 signals are critical for controlling the function and differentiation of Foxp3+ regulatory T cells. OX40-OX40L interaction regulates T-cell tolerance, peripheral T-cell homeostasis, and T-cell-mediated inflammatory diseases.

·第一链cDNA合成试剂盒(预混试剂)(去除基因组DNA)

  本试剂盒是以mRNA或者总RNA为模板，高效合成第一链cDNA。本产品含有反转录反应所需的全部试剂（BalbScript Reverse Transcriptase，RNase Inhibitor，Oligo(dT)18，Random N6，dNTPs），浓度为2×。反应时只需要加入RNA模板和水即可，操作简单，降低操作过程中的污染。本制品含有去除基因组的成分gDNA Purge，可以同步去除基因组DNA污染。反应结束后，85℃加热5秒钟，就可以失活反转录酶、RNA酶抑制剂以及gDNA Purge。合成的第一链cDNA能直接用于PCR或荧光定量PCR的模板以及第二链cDNA的合成或线性RNA扩增的模板，也可用于带有放射性或非放射性核苷酸标记第一链cDNA的实验。

注意事项：
1、用DEPC处理实验用到的所有器材，或者购买已经证明无核酸酶的实验用具，整个实验过程需戴手套并经常更换手套，避免RNA酶的污染。
2、确保所用试剂中无RNA酶污染。
3、试剂盒如不使用，要严格密封保存。在反转录过程中，所有管盖要确保扣严。
4、纯化过的RNA要保证不含有盐，金属离子，乙醇和*酚，因为以上成分会干扰第一链cDNA的合成反应。可采用乙醇沉淀法去除痕量污染物。

实验步骤：

第一链cDNA合成:
1、融化后，将试剂盒中各组分混匀并稍微离心后置于冰上。
2、按顺序加入下面的反应物

模板RNA	总RNA(0.1 ng -5μg) poly(A) mRNA(10 pg) 特异性RNA(0.01 pg)
2×BalbScript PreMix UltraMix	10μl
gDNA Purge	1μl
RNase Free Water	至20μl

3、可选优化步骤。如果RNA模板GC含量高或者含有二级结构，可先将RNA模板和RNase Free Water混匀后，65℃孵育5分钟，冰上冷却后再加入其它组分。
4、轻柔混匀后离心，于42℃条件下孵育60分钟。 如果RNA模板中不含poly A结构，可先在25℃条件下孵育5分钟，随后转到42℃条件下孵育60分钟。
5、70℃加热5min终止反应。
反应产物可直接用于PCR反应，如果不立即使用，在-20℃可保存一周左右。如想延长保存时间，建议使用-70℃保存。

第一链cDNA的PCR扩增:
合成的第一链cDNA可直接用于PCR或qPCR。第一链cDNA反应液体积不能超过PCR反应体系的1/10。通常50 μl的PCR反应体系中，第一链cDNA反应液加入2μl。
