关于Trp A：
Tryptophan synthase is an enzyme that catalyzes the final two steps in the biosynthesis of tryptophan. It is commonly found in Eubacteria, Archaebacteria, Protista, Fungi, and Plantae, but is absent from animals such as humans. Tryptophan synthase typically exists as an α-ββ-α complex.The alpha subunit is responsible for the aldol cleavage of indoleglycerol phosphate to indole and glyceraldehyde 3-phosphate: L-serine + 1-C-(indol-3-yl)glycerol 3-phosphate = L-tryptophan + D-glyceraldehyde 3-phosphate + H2O.The beta subunits catalyze the irreversible condensation of indole and serine to form tryptophan in a pyridoxal phosphate (PLP) dependent reaction. Their assembly into a complex leads to structural changes in both subunits resulting in reciprocal activation.

关于DAN：
Differential screening-selected gene aberrative in neuroblastoma (DAN) is a member of the DAN family of secreted glycoproteins. DAN family antagonists are characterized by a DAN domain that contains a cystine knot motif which is essential for binding to BMP ligands. Members of this family include DAN, gremlin, protein related to DAN and cerberus (PRDC), cerberus, sclerostin (SOST) and uterine sensitization-associated gene 1 protein, and control diverse processes in growth, development and the cell cycle. It has also been reported that DAN family plays crucial role in early mouse embryo development by inhibiting the action of bone morphogenic proteins and modulating the action of transforming growth factor-β superfamily members. DAN is synthesized by small-to intermediate-sized DRG neurons and transported to the sensory nerve terminals in the skin or to the sensory nerve terminals in the dorsal horn. It has been reported that DAN is ubiquitously expressed in adult rat and human tissues. Morphological studies have revealed that, in adult rat, DAN mRNA is expressed ubiquitously in lung and brain, but not in liver.

关于SPARC：
Secreted Protein Acidic and Rich in Cysteine (SPARC) is a secreted, evolutionarily conserved collagen-binding glycoprotein and belongs to the SPARC family. SPARC has 286 amino acids and contains an EF-hand in C-termina domain, a follistatin-like domain with Kazal-like sequences. There are two calcium binding sites, one binds 5 - 8 Ca2+ with a low affinity and other on an EF-hand loop that binds a Ca2+ ion with a high affinity. It is highly expressed in tissues undergoing morphogenesis, remodeling and wound repair. SPARC regulate cell growth through interactions with the extracellular matrix (ECM) and cytokines. SPARC bind to numerous proteins of the ECM, affect ECM protein expression, influence cellular adhesion and migration, and modulate growth factor-induced cell proliferation and angiogenesis. SPARC also binds several types of collagen, albumin, thrombospondin, PDGF and cell membranes.
