hetingcopy 大家好,我现在正在做慢病毒包装,用的是CLONTECK的包装系统,第一次感染293T细胞后,荧光很亮,48H后收集病毒上清,离心后冻于-80度,一天后感染靶细胞,什么都没有?请问大家是怎末做的?谢谢。 大鹏鸟 看到荧光并不代表包装成功,它代表的只是你的质粒转进去了。做慢病毒包装时必须要搭配好几个质粒的比例。转染之后要能看到细胞形态的病变。收集上清的时候要注意尽快分装保存。有可能的话最好把细胞也裂解一下
