5× PCR EnhancerII能与所有的耐热DNA聚合酶共同作用促进许多DNA模板的有效扩增，增加PCR反应的灵敏度和特异性。在PCR反应中，PCR增强剂主要通过提高DNA聚合酶的热稳定性、降低DNA的二级结构的含量来发挥作用，因此在遇到一些复杂模板(如GC含量高)，或具有重复序列的模板时，可通过提高退火温度，极大地减少DNA二级结构对PCR的影响，同时对酶的活性没有改变。
